It could work as a multiplayer of 100 players with the same map but doing combos. Whoever makes the highest combo wins and the other player is disqualified. It would be interesting to see this style of play in osu!
Although it would be difficult to play with 100 people at the same time if you have a slow connection, it would be better to do it as a 1vs1 tournament and there would also be rewards for the winners, it could be PP or medals.
