So I was born left handed and when I was younger I'd go on the computer and the mouse would be on my right side, So I'd learnt to use the mouse with my right hand, Now obviously with me holding a pen in my left hand all my life, and learning to use buttons X/Z with my Left hand has anybody had the same experience and would shed some light about what they did?

I thought so going to be weird getting used to though now to just buy my tablet :p
Plenty of lefties hold the tablet's pen in their left hand. >.> Just go with what you prefer.
I switched from right-handed mouse to left-handed tablet. The difficulty in switching hands isn't the pen (it should feel natural after a small amount of time), it's in the keyboard tapping. If you built any amount of speed playing right-handed (and don't play other rhythm games that require both hands, like Stepmania), then be prepared to rebuild that speed from the very beginning.
