Well, definitely Gintama, especially if you have some knowledge about other animes. Also, Soul Eater was quite funny in my opinion.

I have watched a number of anime comedies but Gintama is the only one that has managed to give me stomach cramps almost every episode so far.

Funniest anime I've seen HAS to be Carnival Phantasm, although you'll have to watch Tsukihime and Fate/stay night beforehand to completely get it.
You can understand most of it with just Fate/stay night, like I've seen Fate/stay night but not Tsukihime, still, I thoroughly enjoyed it. Don't watch it if you haven't seen either of the two, you will end up more confused than entertained.
