I received a mod on my map asking to set combo colors for all diffs. It appears that I have 4 combo colors and I'm not sure how to check if they are set. Can someone enlighten me on this topic?

If you combo colours look like the above image, they probably aren't set since those are the defaults. You can verify this by looking at the .osu file; if the combo colours are set, there's a section titled [Colours] between [TimingPoints] and [HitObjects].
To set the colours, click on the colours in Song Setup and pick whatever colours you like. Conventionally, people pick combo colours that match the background.
