I'm not getting any error, the little hourglass beside the mouse cursor appears for a few seconds, then disappears.
I tried re-installing with the install that doesn't require .net frameworks 2.0 and it's still not working.

It keeps testing the login or something, but never actually opens the game O_o
I use dial up connection. it's slow, but I tought it was supposed to work, since it shouldn't download anything unless I asked, I belive.

It's working now that I updated. It's just uber laggy. 
I had played osu! for a few months already
Today i download a few new song and click on osu! exe for update, i found that i can't run it!
And i got the error message
Firstly i thought it was the problem of .Net Framework problem
But i try uninstall and reinstall it problem still can't solved
than i try reinstall osu as well

If it's helpful to anyone. I had the initialization error yesterday and could not figure out why.
Well eventually I realized that deleting my osu!.cfg file would fix it. I was curious to why this was, and it turns out if your currently selected skin's skin.ini has a NULL value, osu! will not start. I don't really know how that happened in the first place, but it could happen to you too.
Example:

It happens that I have started getting this error after I have changed some settings on the audio in the control panel (I'm using windows 7). I tried it on another computer (also Windows 7). I can play it at first, but after I clicked Control Panel>Hardware and Sound>Audio >Cancel (I didn't change anything) then Osu! is suddenly unplayable with the above error.
Is it because of the uncompatability with windows 7?

I guess you tried everything but searching. This is a simple one - uninstall XNA game studio. Microsoft fucked up with the GAC resolution for it and you it is not playing nice with osu!. This only happens if you have an old old version, anyway.Melon Bread wrote:
